Prothrombin activation by a metalloprotease from Staphylococcus aureus.

چکیده
Formation of thrombin during incubation of purified bovine prothrombin with purified staphylococcal metalloprotease has been investigated. Thrombin activity was estimated by examination of clotting time and by digestion of a synthetic substrate, Chromozym TH. The metalloprotease caused direct activation of prothrombin which was inhibited by the addition of ethylenediaminetetraacetic acid. Metalloprotease produced by some strains of Staphylococcus aureus may simulate staphylocoagulase activity.

Dabigatran inhibits Staphylococcus aureus coagulase activity.
The ability of Staphylococcus aureus to clot plasma through conformational activation of prothrombin by staphylocoagulase is used to distinguish S. aureus from coagulase-negative staphylococci. We show that while the direct thrombin inhibitor dabigatran inhibits staphylocoagulase activity, the clinical use of dabigatran etexilate is not expected to interfere with direct tube coagulase testing.
متن کاملThe human fibrinolytic system is a target for the staphylococcal metalloprotease aureolysin.
The major opportunistic pathogen Staphylococcus aureus utilizes the human fibrinolytic system for invasion and spread via plasmin(ogen) binding and non-proteolytic activation. Because S. aureus secretes several proteases recently proposed as virulence factors, we explored whether these enzymes could add to the activation of the host's fibrinolytic system. Exposure of human pro-urokinase [pro-uP...
متن کاملComparison of the affinities to bovine and human prothrombin of the staphylocoagulases from Staphylococcus intermedius and Staphylococcus aureus of animal origin.
Staphylocoagulases of Staphylococcus aureus (40 strains originally isolated from horses, dogs, cats, cows, sheep, opossums, pigs, humans, and a goat) and Staphylococcus intermedius (19 isolates from dogs and 1 pigeon strain) were tested for their affinity to prothrombins of either bovine or human origin. The tests used were the coagulase tube test (using human, bovine, or equine fibrinogen with...
متن کاملContribution of Coagulases towards Staphylococcus aureus Disease and Protective Immunity
The bacterial pathogen Staphylococcus aureus seeds abscesses in host tissues to replicate at the center of these lesions, protected from host immune cells via a pseudocapsule. Using histochemical staining, we identified prothrombin and fibrin within abscesses and pseudocapsules. S. aureus secretes two clotting factors, coagulase (Coa) and von Willebrand factor binding protein (vWbp). We report ...
متن کاملAdhesion of Staphylococcus aureus to the vessel wall under flow is mediated by von Willebrand factor-binding protein.
Adhesion of Staphylococcus aureus to blood vessels under shear stress requires von Willebrand factor (VWF). Several bacterial factors have been proposed to interact with VWF, including VWF-binding protein (vWbp), a secreted coagulase that activates the host's prothrombin to generate fibrin. We measured the adhesion of S aureus Newman and a vWbp-deficient mutant (vwb) to VWF, collagen, and activ...
